Genistein powder Production Process
1.Preparation of raw material
Firstly, select dry, mold-free, and qualified Sophora japonica. Next, the dried Sophora japonica are rinsed with clean water to remove impurities such as sand and mud. Then, the cleaned Sophora japonica are dried and crushed them into coarse or fine powder using a grinder.
2.Extraction
Ethanol aqueous solution (such as 60%-90% ethanol) is commonly used as the extraction solvent. Ethanol is safe and moderately priced, has good solubility for isoflavones, and is easy to recover.
Place the Sophora japonica powder and a certain proportion of ethanol solution in a reflux extraction tank, heating to boiling, and maintain for a certain period of time (3 hours).
3.Solid-liquid separation
After each extraction, separate the extract from the residue while it is still hot (to prevent the loss of active components due to cooling and precipitation or the precipitation of impurities). Then, combine all the filtrates and discard the residue.
4.Concentration of extract
Using a rotary evaporator or vacuum concentrating tank at a relatively low temperature (50-65℃), recover most of the solvent (ethanol) from the extract to obtain a concentrated extract.
5.Purification
The concentrated extract typically contains a large amount of water-soluble impurities (such as sugars, proteins, pigments) and lipid-soluble impurities.
Adopt the method of Macroporous Adsorption Resin, first, dilute the concentrated extract to an appropriate concentration with water and load it onto a pretreated Macroporous Adsorption Resin column.
Rinse the resin column with a large amount of water to wash away water-soluble impurities (such as sugars, salts, proteins); Then elute with an ethanol aqueous solution of a certain concentration (30%-70%), Genistein is eluted at this stage. The eluate fractions containing Genistein are collected in segments.

6.Refinement and drying
The collected elution rich in Genistein is concentrated to a small volume or into an extract from under reduced pressure. Sometimes, recrystallization using an appropriate solvent (ethanol) is performed to further improve purity.
The concentrated Genistein solution or extract is dried to obtain crude or purified Genistein. Drying is carried out under reduced pressure at a low temperature to avoid thermal decomposition.
If the purity is aceptable and a powder form is desired, the concentrated solution can be directly spray-dried, but attention should be paid to the potential impact of high temperature. Finally, the dried block is crushed and sieved to obtain uniform Genistein powder.
